There is a fair chance one of my teams is going to lock up the #1 seed with 2 or 3 games to go in the season. That could potentially give some of my studs 4 games off, 3 plus a bye.
Aside from potentially losing some conditioning, any negatives to doing this? Do players get rusty? :) I just want to avoid injury more than anything

I have never noticed a player to be rusty after a couple games off. They will just lose some conditioning the more they sit. 1-2 games wouldn't hurt too bad. 3-4 games plus a bye week for playoffs might be a bit much. You might give a little more time to the players with those 3-4 game injuries to make sure they are ready for playoffs. Just keep an eye on their conditioning (on player card) after every game.
If you are talking about your Chicago team in MFN-20, please sit all starters this week. You are playing my New Orleans team and I need a win! ;)
